This guide was made specifically for Leica programs and microscopes. The microscope we used to create this how-to guide is the Leica M205 C, consisting of z-stacking capabilities and a motorized column for effortless imaging.
Iris Diaphragm
The Iris Diaphragm regulates the available light, which changes the depth of field. The depth of field is the area of a specimen that is brought into sharp focus.
Start with it open (all the way to the left). The specimen will appear brighter and in higher resolution, but the depth of field will decrease.
When it is turned to the right, the size of the aperture decreases. This makes the specimen appear darker and lowers the resolution, but increases the depth of field. This can be useful when users struggle to get a specimen in complete focus. Only try it if changing the contrast and lighting does not work.
