This guide was made specifically for Leica programs and microscopes. The microscope we used to create this how-to guide is the Leica M205 C, consisting of z-stacking capabilities and a motorized column for effortless imaging.
Objectives
Currently, the Seed Conservation Lab has two plan-apochromatic objectives, the 0.63x and the 1.6x, that can be used to image specimens.
Plan-apochromatic describes an apochromatic lens system that produces a planar, or flat, field. An apochromatic lens system means the lens is corrected for chromatic and spherical aberration.
The 0.63x objective can be utilized for larger specimens, such as Zanthoxylum and Dubautia, while the 1.6x can be utilized for smaller specimens, such as Liparis or Cyrtandra.
The largest specimen that can be placed under the 1.6x objective is 5 mm wide. If you are imaging seeds greater than that size, switch to the 0.63x objective lens.
The largest specimen that can be placed under the 0.63x objective is 4 mm in width and 11.5 mm in length (subject to change).
Changing the Objectives
ALWAYS wear gloves when changing objectives to prevent the lens from getting dirty. Use the handwheel to focus OUT to raise the focusing column before switching objectives. This will ensure you have plenty of room to handle the objectives comfortably without causing damage.
The 0.63x objective needs the O-ring attachment. It can be found with the lens.
Loosen the dome by turning the black knob in the center to the left.
Once it slides off with ease, the objective can be seen.
Turn the objective to the left until it comes off the track.
Users must handle the objective with two hands. It is heavier than expected, so do not let it crash into the stage.
Put the lens in its designated box, and screw the new lens in. Turn it to the right until it is tight, slide the dome over it, and tighten it by turning the knob to the right. Double-check that the dome is on tight; it will slide down if it is not.
You must update the software in LAS X once you switch the objectives.

If the clamps are changed, that needs to be indicated when opening up the Leica program, by choosing ‘Yes’ when prompted for the Z-Drive.
An Alan wrench is needed to move the clamps.
