This guide was made specifically for Leica programs and microscopes. The microscope we used to create this how-to guide is the Leica M205 C, consisting of z-stacking capabilities and a motorized column for effortless imaging.

To Stamp An Image

Once you create annotations or a scale bar, you must ensure they appear on the overlay. To do so, click on the Show or Hide Individual Overlays button at the top left of your screen.

It will look something like this:

This window allows you to activate or deactivate the display of certain functions.

When a red eye is visible on a certain function (in this case, the annotations and the scale bar), they will be displayed over the image. To deactivate, press those buttons.

The stamp symbol allows you to automatically burn in annotations, measurements, and the scale bar directly into the image. When it is selected, the stamp icon should appear at the bottom left of your screen.

Note: If you are in Live Image Mode, you will not see a stamp but a blue icon where the camera icon used to be. The stamp will appear once you are out of Live Mode, i.e., when you click on the pause button or on an image you took.

Once you create a Z-stacked image, it will appear in the Explorer page as a folder.

Inside the folder, click on Multifocus Image – 1. This is the final Z-stacked image.

You can now use the stamp button (highlighted) to burn the scale bar onto the chosen image.

This will create a new photo labeled ‘Multifocus Image-1_Overlay’. It will show the scale bar as an overlay on the image.

Note: if you made any other annotations on the image, they will be stamped on unless they are unselected in the Show/Hide Individual Overlays window.

You can then rename the image by right-clicking on it.
