This guide was made specifically for Leica programs and microscopes. The microscope we used to create this how-to guide is the Leica M205 C, consisting of z-stacking capabilities and a motorized column for effortless imaging.
Focus Range
The first step is to ensure that the ‘Begin’ and ‘End’ on the z-stacking settings are set at the end of both focal ranges.
This means that when users set each ‘Begin’ and ‘End,’ the specimen should be out of focus. When the range is set this way, the camera should be able to capture the full range of the specimen.
Note that setting the ‘Begin’ and ‘End’ too far out of focus can also create issues, as the camera will pick up the background as part of the z-stack. Use the fine focus adjustments on the software if needed to set your focus.
